Iovance Biotherapeutics is accelerating the expansion of its tumor-infiltrating lymphocyte (TIL) therapy platform across a broader range of solid-tumor indications following a second quarter that generated nearly $100 million in revenue and a 56% margin. The company is transitioning its proprietary TIL technology from academic research settings into a commercial model for personalized cancer treatment.

Brian Gastman, executive vice president of translational medicine and research at Iovance, confirmed the company has now treated more than 2,000 patients through clinical and commercial channels. The firm currently maintains two commercialized therapies, Amtagvi and the interleukin-2 treatment Proleukin, as it seeks to increase its market share in the oncology sector.

The manufacturing process relies on the Iovance Cell Therapy Center in Philadelphia, which serves as a central hub for expanding patient-specific immune cells into the billions. Gastman noted that the facility possesses modular capacity to support more than 5,000 patients, providing the operating leverage necessary to manage increasing treatment volumes.

Market access remains a primary focus for the company, which currently operates more than 95 authorized treatment centers across the United States. With roughly one-third of these locations situated in community settings, the company aims to reach 110 centers by the end of the year to improve patient proximity.

Payer coverage has expanded to include more than 250 million lives, encompassing both Medicare and commercial insurance plans. This infrastructure covers over 75% of the U.S. population, ensuring that the majority of patients reside within 200 miles of a facility capable of administering the specialized therapy.

Clinical data for Amtagvi in heavily pretreated melanoma patients continues to demonstrate durable responses, according to company analysis. In a review of four treatment centers, Iovance reported a 44% response rate, with efficacy exceeding 50% among patients receiving the therapy in the second-line setting.

The company is now evaluating TIL therapy in earlier stages of advanced melanoma through the IOV-COM-202 Cohort 1A trial. Data from the combination of Amtagvi and pembrolizumab showed response rates in the mid-60% range, with a 30% complete-response rate among immune checkpoint inhibitor-naïve patients.

Beyond melanoma, Iovance is targeting non-small cell lung cancer, soft-tissue sarcoma, and endometrial cancer with its standard TIL platform. The lung cancer program, which has received Fast Track designation from the FDA, reported a response rate of nearly 26% with sustained duration of response in early follow-up data.

The company is also diversifying its portfolio with next-generation programs, including genetically modified TIL therapies and engineered interleukin-2. These initiatives, such as the PD-1-inactivated IOV-4001 and the IL-12-expressing IOV-5001, aim to address tumors that have historically proven difficult to treat with standard immunotherapy approaches.

The strategic shift toward these engineered therapies reflects an effort to limit systemic exposure while maximizing the impact of the treatment within the tumor microenvironment. By focusing on indications like colorectal and triple-negative breast cancer, Iovance intends to broaden the utility of its cell therapy platform and address unmet clinical needs in complex solid tumors.

Furthermore, the company is actively refining its manufacturing protocols to reduce production turnaround times, which remains a critical factor in patient outcomes for autologous cell therapies. This operational efficiency is expected to support the company’s long-term commercial viability as it transitions from a clinical-stage developer to a commercial-stage leader.

Future milestones include the presentation of more mature data from the IOV-COM-202 trial at the upcoming European Society for Medical Oncology meeting. The company also expects to complete its current lung cancer trial and submit a supplemental biologics license application in 2027.

The international Phase III TILVANCE-301 confirmatory trial remains a critical watchpoint for the company. Successful outcomes in this study are expected to support the confirmation of the company’s second-line accelerated approval and potentially establish a path for frontline use.
